ฤทธิ์ของสารสกัดสมุนไพรรางจืดและลูกใต้ใบในการลดการหลั่ง IL-1 จากเซลล์ LPS-Activated THP-1 Macrophages เพาะเลี้ยง

บทคัดย่อ

การอักเสบเป็นการตอบสนองขั้นพื้นฐานของร่างกายเพื่อป้องกันสิ่งที่อาจเป็นอันตราย หากเกิดขึ้นอย่างไม่เหมาะสมหรือควบคุมไม่ได้ ย่อมก่อให้เกิดความเสียหายต่อเซลล์และเนื้อเยื่อ และนาไปสู่พยาธิสภาพการเกิดโรคเสื่อมต่างๆ ยาต้านอักเสบที่นิยมใช้กันอย่างแพร่หลายในปัจจุบันได้แก่ยาในกลุ่ม Nonsteroidal anti-inflammatory drugs (NSAIDs) ซึ่งมีผลข้างเคียงมากโดยเฉพาะต่อระบบทางเดินอาหารและระบบหัวใจหลอดเลือด ทาให้มีการพัฒนาและค้นคว้าหาสารต้านอักเสบจากธรรมชาติเพื่อมาใช้ทดแทน การวิจัยครั้งนี้มีวัตถุประสงค์เพื่อศึกษาฤทธิ์ของสารสกัดรางจืดและลูกใต้ใบที่มีต่อพฤติกรรมของเซลล์ macrophage like-THP-1 ในการหลั่งไซโตไคน์ IL-1 โดยจะทาการเปรียบเทียบปริมาณ IL-1 ที่หลั่งจากเซลล์เมื่อทดสอบเซลล์กับสารสกัดรางจืดความเข้มข้น 200-800 μg/ml สารสกัดลูกใต้ใบความเข้มข้น 200-800 μg/ml สารต้านอักเสบอ้างอิง 2 ชนิดได้แก่ Epigallocatechin gallate (EGCG) (50 nM) และ IL-10 (10 ng/ml) ในสภาวะทดสอบร่วมกับสารก่ออักเสบ LPS (1,000 ng/ml) เป็นเวลา 24 ชั่วโมง ผลการทดสอบพบว่า เมื่อเซลล์ถูกกระตุ้นด้วย LPS จะหลั่ง IL-1 เพิ่มขึ้นอย่างมีนัยสาคัญทางสถิติ (p=0.000) จากนั้นเมื่อทาการทดสอบร่วมระหว่าง LPS กับสารสกัดสมุนไพรพบว่าสารสกัดรางจืดความเข้มข้น 400-800 μg/ml และสารสกัดลูกใต้ใบความเข้มข้น 200-800 μg/ml สามารถยับยั้งการหลั่ง IL-1 ที่เกิดจากการกระตุ้นด้วย LPS ได้อย่างมีนัยสาคัญทางสถิติ (p=0.000) โดยการยับยั้งของสารสกัดสมุนไพรทั้ง 2 ชนิดเป็นไปในลักษณะ dose- dependent manner อย่างไรก็ตาม ในการศึกษานี้ EGCG (50 nM) และ IL-10 (10 ng/ml) ซึ่งเป็นสารต้านอักเสบอ้างอิงไม่สามารถยับยั้งการหลั่ง IL-1 จาก THP-1 ที่กระตุ้นด้วย LPS ความเข้มข้น 1,000 ng/ml ได้อย่างมีนัยสาคัญทางสถิติ จากผลการศึกษานี้แสดงให้เห็นว่า สารสกัดรางจืดและสารสกัดลูกใต้ใบมีสารออกฤทธิ์ต้านการอักเสบที่อาจนาไปพัฒนาไปเป็นยาต้านการอักเสบชนิดใหม่จากธรรมชาติได้ในอนาคต
