การประยุกต์ใช้วิธีพื้นผิวตอบสนองเพื่อทำนายสภาวะที่เหมาะสมในการลดการเกิดสีน้ำตาลของชิ้นส่วนต้นกล้ากล้วยไข่ในสภาพปลอดเชื้อ

ปัญหาที่สำคัญประการหนึ่งของการเพาะเลี้ยงเนื้อเยื่อกล้วยไข่ คือ ปัญหาการเกิดสีน้ำตาลจากการออกซิเดชันของสารประกอบฟีนอลิก การแก้ไขปัญหาทำได้โดยเติมสารต้านอนุมูลอิสระหรือสารดูดซับฟีนอลิก การศึกษานี้มีวัตถุประสงค์เพื่อหาสภาวะที่เหมาะสมสำหรับลดการเกิดสีน้ำตาลของชิ้นส่วนเพาะเลี้ยงโดยการใช้วิธีพื้นผิวตอบสนอง (response surface methodology) กล่าวคือ การสร้างแบบจำลองสภาวะด้วยวิธีการวางแผนการทดลองแบบวิธีพื้นผิวตอบสนองร่วมกับการวางแผนการทดลองแบบเซ็นทรัลคอมโพสิตของปัจจัย 3 ปัจจัยคือ ascorbic acid (AA) polyvinylpyrrolidone (PVP) และ activated charcoal (AC) ความเข้มข้น 3 ระดับ จำนวน 17 ทรีตเมนต์ เป็นเวลา 12 สัปดาห์ เพื่อนำค่าของปริมาณฟีนอลิก แทนนิน และฟลาโวนอยด์ของอาหารเพาะเลี้ยงเนื้อเยื่อพืชที่มีค่าน้อยที่สุด เพื่อสร้างแบบจำลองสภาวะที่เหมาะสมอีก 4 ทรีตเมนต์ จากนั้นทดสอบแบบจำลอง เป็นเวลา 12 สัปดาห์ ผลการทดลองพบว่าสภาวะของแบบจำลองที่เหมาะสมคือ อาหาร Murashige & Skoog (1962) ที่เติม AA 0.03 กรัมต่อลิตร ร่วมกับ PVP 3.325 กรัมต่อลิตร และ AC 1.48 กรัมต่อลิตร สามารถลดการเกิดสีน้ำตาลในการเพาะเลี้ยงต้นกล้ากล้วยไข่ในสภาพปลอดเชื้อได้อย่างมีประสิทธิภาพ
