EFFECT OF THE CRUDE PIGMENT EXTRACT FROM ACTINOBACTERIA ISOLATED FROM STINGLESS BEE, TETRAGONILLA COLLINA (SMITH, 1857), ENTRANCE NEST ON NATURAL DYEING FIBERS

Abstract

Actinobacterial dyes are the alternative dyes for natural fiber dyeing. In this study, the C4 strain isolated from stingless-bees hive was evaluated for using as the dyes in textile dyeing process. Morphologically, the C4 colonies grown on sodium caseinate agar (SCA) produced the water-soluble violet-colored pigment. To identify the actinobacterial C4 strain, phylogenetic identification based on the partial 16S rDNA sequence analysis was determined. The result showed that the strain was identical compared with Streptomyces californicus (100% sequence identity). In order to upscale of actinobacterial dye production, the C4 strain was cultivated on the media supplemented with the broken-milled rice as the carbon and energy sources. After the actinobacterial growth, the pigment produced was extracted by 70 % (v/v) ethanol in order to obtain the actinobacterial dye. A total of 5 natural fibers, including silk, wool, cotton fiber, hemp fiber, and the mixed banana and cotton fibers, was soaked in the actinobacterial dyes extracted. After 12 h of dyeing, the color measurement followed by CIELAB system and the Standard Color Chart R.H.S were applied with all natural fibers. The dyeing results indicated that silk showed the highest CIELAB value indicated that it was the best natural fibers suitable for actinobactrial dyeing. The use of actinobacterial dye in textile industry should be evaluated in future.The dyeing results indicated that silk showed the highest CIELAB value indicating that it is the most suitable natural fiber for actinobacterial dyeing. It is suggested that the use of actinobacterial dye in the textile industry should be evaluated in future.
