การวิเคราะห์ความสัมพันธ์ทางพันธุกรรมของไผ่ 12 ชนิดในจังหวัดเพชรบูรณ์ โดยเครื่องหมายโมเลกุล ISSR

บทคัดย่อ
การศึกษานี้มีวัตถุประสงค์เพื่อวิเคราะห์ความสัมพันธ์ทางพันธุกรรมของไผ่ 12 ชนิด โดยใช้เครื่องหมายโมเลกุล ISSR (Inter-Simple Sequence Repeats) เพื่อสร้างลายพิมพ์ดีเอ็นเอและวิเคราะห์ความใกล้ชิดเชิงวิวัฒนาการของชนิด จากการสกัดดีเอ็นเอจากตัวอย่างใบไผ่จำนวน 36 ตัวอย่าง ด้วยชุดสกัดสำเร็จรูป RBC Genomic DNA Kit Genomic DNA Extraction Mini Kit บริษัท RBC Bioscience ดีเอ็นเอที่ได้มีปริมาณเฉลี่ย 6.5 นาโนกรัมต่อไมโครลิตร มีค่าความบริสุทธิ์เฉลี่ยที่ 1.8 ทำปฏิกิริยา PCR ด้วยเครื่องหมายโมเลกุล ISSR จำนวน 5 เครื่องหมาย โดยเครื่องหมาย UBC807 ให้ผลแถบดีเอ็นเอชัดเจนที่สุด วิเคราะห์ค่าดัชนีความคล้ายคลึง และใช้วิธี UPGMA เพื่อสร้าง dendrogram ผลการวิเคราะห์พบว่า ค่าสัมประสิทธิ์ความคล้ายคลึงอยู่ระหว่าง 0.63 - 1.00 และสามารถจำแนกไผ่ทั้ง 12 ชนิดออกเป็น 5 กลุ่มหลัก ได้แก่ กลุ่มที่ 1 ไผ่ซาง ไผ่คะมะ และไผ่บงดำ กลุ่มที่ 2 ไผ่บง ไผ่จีน และไผ่หลอด กลุ่มที่ 3 ไผ่เป๊าะ ไผ่ไร่ และไผ่น้ำเต้า กลุ่มที่ 4 ไผ่สีสุกและไผ่เหลือง และกลุ่มที่ 5 ไผ่คายดำ ผลการจำแนกกลุ่มดังกล่าวสะท้อนความหลากหลายทางพันธุกรรมและความใกล้ชิดระหว่างชนิด โดยบางกลุ่มมีค่าสัมประสิทธิ์สูงกว่า 0.80 ซึ่งแสดงถึงความใกล้เคียงกันมาก ในขณะที่บางกลุ่ม เช่น ไผ่คายดำ แสดงการแยกตัวชัดเจน ผลการศึกษานี้จึงชี้ให้เห็นถึงศักยภาพของการใช้เครื่องหมายโมเลกุล ISSR ในการศึกษาความสัมพันธ์เชิงพันธุกรรมของไผ่ ซึ่งเป็นประโยชน์ต่อการปรับปรุงพันธุ์ การอนุรักษ์ความหลากหลายทางชีวภาพ และการใช้ประโยชน์ทางเศรษฐกิจของไผ่ต่อไป
